چکیده

The joining of the 15-carbon farnesyl group ðC15H25Þ and the 20-carbon geranylgeranyl group ðC20H33Þ to protein–cysteines at or near their carboxy-termini is catalyzed by protein farnesyltransferase (FTase) and protein geranylgeranyltransferase-I and II (GGTase-I and GGTase-II) [1]. The prenyltransferases are heterodimers consisting of aand b-subunits with combined molecular masses ranging from 91 to 98 kDa. The asubunits of FTase and GGTase-I are the same, and the b-subunits differ. The b-subunits of the three enzymes are homologous to the a-subunits and to each other. The overall reactions are shown by the following chemical equations: FPPþHS-peptide ! farnesyl-S-peptideþ PP GGPP þHS-peptide ! geranylgeranyl-S-acceptor þ PP

Protein prenylation in spinach chloroplasts.

Protein prenylation in plants was studied by in vivo metabolic (3)H-mevalonate labeling in combination with a range of protein synthesis inhibitors. In spinach cotyledons, this posttranslational protein modification was found to be divided into two categories, one representing the conventional prenylation involving farnesyl and geranylgeranyl groups bound to cysteine residues via thioether link...

Increased RhoA Prenylation in the loechrig (loe) Mutant Leads to Progressive Neurodegeneration

The Drosophila mutant loechrig (loe) shows age-dependent degeneration of the nervous system and is caused by the loss of a neuronal isoform of the AMP-activated protein kinase (AMPK) γ-subunit (also known as SNF4Aγ). The trimeric AMPK complex is activated by low energy levels and metabolic insults and regulates multiple important signal pathways that control cell metabolism. A well-known downst...

Relative Contributions of Prenylation and Postprenylation Processing in Cryptococcus neoformans Pathogenesis

Prenyltransferase enzymes promote the membrane localization of their target proteins by directing the attachment of a hydrophobic lipid group at a conserved C-terminal CAAX motif. Subsequently, the prenylated protein is further modified by postprenylation processing enzymes that cleave the terminal 3 amino acids and carboxymethylate the prenylated cysteine residue. Many prenylated proteins, inc...

RhoB prenylation is driven by the three carboxyl-terminal amino acids of the protein: evidenced in vivo by an anti-farnesyl cysteine antibody.

Protein isoprenylation is a lipid posttranslational modification required for the function of many proteins that share a carboxyl-terminal CAAX motif. The X residue determines which isoprenoid will be added to the cysteine. When X is a methionine or serine, the farnesyl-transferase transfers a farnesyl, and when X is a leucine or isoleucine, the geranygeranyl-transferase I, a geranylgeranyl gro...

Regulation of a Drosophila melanogaster cGMP-specific phosphodiesterase by prenylation and interaction with a prenyl-binding protein.

Post-translational modification by isoprenylation is a pivotal process for the correct functioning of many signalling proteins. The Drosophila melanogaster cGMP-PDE (cGMP-specific phosphodiesterase) DmPDE5/6 possesses a CaaX-box prenylation signal motif, as do several novel cGMP-PDEs from insect and echinoid species (in CaaX, C is cysteine, a is an aliphatic amino acid and X is 'any' amino acid...
